    The Effect of Surfactants and pH Modifying Agents on the Dissolution and Permeation of Pimobendan

    Solubility and permeability are key parameters for establishing in vitro-in vivo correlation for poorly water-soluble active pharmaceutical ingredients (APIs). Recent studies demonstrate that not only solubility, but also effective permeability of the API may change due to the addition of solubilizing agents, and there is a certain mathematical relation between these physicochemical parameters. The aim of this study was to show the importance of early screening of solubility and permeability in presence of additives in order to achieve the expected bioavailability of the API. In this work, the effect of surfactants and microenvironmental pH modifiers were in focus, and pimobendan was chosen as model drug.In the case of pH modifiers, the equilibrium solubility of the API increased, while the permeability decreased significantly. No negative effect was observed for two surfactants at low additive levels, but these two additives also exhibited a slightly negative effect on permeability when used at higher concentrations. In the simultaneous dissolution-permeation studies the surfactants-containing formulation was found to have slightly higher flux than the pH-modifier-containing one. It can be due to the phenomenon that the dissolution of the active substance can be enhanced by these surfactants without any significant permeability reducing effect.The results obtained from the present study clearly demonstrate the importance of studying drug-additive interactions in every step of formulation development and based on these, the selection of the appropriate quality and quantity of additives. In addition, the results also underline the significance of performing simultaneous dissolution-permeation studies to predict bioavailability

    Promoting SME cooperative aggregations: main criteria and contractual models

    Collaboration is considered an effective solution to improve business strategies. However, small and medium enterprises (SMEs) often lack common principles and common forms of contractual coordination. Several policies implemented by the EU have addressed the set-up of a comprehensive SME policy framework, but European institutions seem to have focused more on organisational devices to conduct business activities rather than on contractual forms of coordination. In April 2009, Italy adopted a law in network contract to promote the development of inter-firm cooperation strategies to foster enterprises' innovation and growth. Even if this law represents a novelty in Europe and may offer new challenges and hints, it still presents some lacks in its formulation. The current research aims at presenting the Italian law for network contract and a comparison with other models of SME aggregations adopted in EU countries. A formal model to support the design of an SME network was proposed, by providing both an ontology-based model to help the definition of the contract in a structured way, and a basic workflow to identify the important phases of the network design, i.e. the feasibility study and the negotiatio

    Association Between Common Cardiovascular Risk Factors and Clinical Phenotype in Patients with Hypertrophic Cardiomyopathy From the European Society of Cardiology (ESC) EurObservational Research Programme (EORP) Cardiomyopathy/Myocarditis Registry

    The interaction between common cardiovascular risk factors (CVRF) and hypertrophic cardiomyopathy (HCM) is poorly studied. We sought to explore the relation between CVRF and the clinical characteristics of patients with HCM enrolled in the EURObservational Research Programme (EORP) Cardiomyopathy registry.1739 patients with HCM were studied. The relation between hypertension (HT), diabetes (DM), body mass index (BMI) and clinical traits was analyzed. Analyses were stratified according to the presence or absence of a pathogenic variant in a sarcomere gene.The prevalence of HT, DM and obesity (Ob) was 37%, 10%, and 21%, respectively. HT, DM and Ob were associated with older age (p<0.001), less family history of HCM (HT and DM p<0.001), higher New York Heart Association (NYHA) class (p<0.001), atrial fibrillation (HT and DM p<0.001; Ob p = 0.03) and LV (left ventricular) diastolic dysfunction (HT and Ob p<0.001; DM p = 0.003). Stroke was more frequent in HT (p<0.001) and mutation-positive patients with DM (p = 0.02). HT and Ob were associated with higher provocable LV outflow tract gradients (HT p<0.001, Ob p = 0.036). LV hypertrophy was more severe in Ob (p = 0.018). HT and Ob were independently associated with NYHA class (OR 1.419, p = 0.017 and OR 1.584, p = 0.004, respectively). Other associations, including a higher proportion of females in HT and of systolic dysfunction in HT and Ob, were observed only in mutation-positive patients.Common CVRF are associated with a more severe HCM phenotype, suggesting a proactive management of CVRF should be promoted. An interaction between genotype and CVRF was observed for some traits

    Fényemittáló maleinimid tartalmú polimerek előállítása és vizsgálata

    Fluoreszcens tulajdonságokkal rendelkező maleimid tartalmú alapláncokat szintetizáltunk. Az előállított monomereket (akril-származékok) gyökös módszerrel polimerizáltuk. A fluoreszcens termékeket tisztítottuk és részletesen karakterizáltuk NMR, GPC és tömegspektrometriás mérésekkel. Az elvégzett optikai vizsgálatok kimutatták, hogy az előállított monomerek és polimerek, az oldat elkészítésénél használt oldószer polaritásától függően, zöld vagy sárga fényt emittálnak
